Abstract

The European eel (Anguilla anguilla) is a catadromous fish with a complicated life cycle. The long-term impact of anthropopressure, environmental pollution and diseases have led to a risk of extinction. The aim of the present study was to determine the influence of Anguillid herpesvirus-1 infection on the innate immunity of European eel from natural conditions. Spleen phagocyte respiratory burst activity and potential killing activity, as well as pronephros lymphocyte proliferation stimulated by concanavalin A or lipopolysaccharide were measured. The analyses of the results showed that all studied parameters were significantly higher (P<0.05) in AngHV-1-negative fish compared to the ones where the presence of viral DNA was confirmed.

Abstract

The aim of the present study was to investigate blood parameters and subsequent fertility in cows with or without increased postpartum polymorphonuclear neutrophil activity. The study was conducted with 15 Brown Swiss cows between 1-3 lactations. Polymorphonuclear neutrophil activities were assessed at 10±4 days before and after parturition. The cows which maintained their phagocytic and oxidative burst activites compared to the prepartum period were classified as control (CON), and cows which increased phagocytic and oxidative burst activites were defined as increased cellular immune response (ICIR) cows. Energy, protein metabolism markers, hepatic enzymes, blood mineral levels and body condition scores were measured at -10±4, 3±2, 10±4 days relative to parturition. Pregnancy rates, the number of inseminations, and calving to pregnancy intervals were evaluated. The mean non-esterified fatty acid (NEFA) and beta -hydroxybutyric acid (BHB) concentrations were lower in ICIR cows. Mean serum calcium (Ca) concentrations were in subclinical hypocalcemia level at day 3±2, 10±4 days postpartum in CON cows. Postpartum immune cell functions and NEFA, BHB concentrations were negatively cor- related. The calving to pregnancy interval were longer in the control cows. However, total preg- nancy rates and the number of insemination in both groups were similar. In conclusion, postpar- tum polymorphonuclear neutrophil activity is affected by periparturient metabolic status. Postpartum energy metabolites negatively affected the postpartum cellular immune response. The increased postpartum polymorphonuclear neutrophil activity at early postpartum period is positively related with subsequent fertility in dairy cows.

Abstract

Florfenicol is a broad-spectrum bacteriostatic antibiotic commonly used for the treatment of systemic infections in farm animals. The aim of this study was to determine the effect of florfenicol on the percentage of T lymphocytes (CD3+, CD4+, CD8+, TCRgd+ cells) and B lymphocytes (Bu-1+ cells) and on total serum anti - sheep red blood cell (SRBC) haemagglutinin titer in the peripheral blood of SRBC–immunized broiler chickens. The study included three groups of broiler chickens differentiated by weight (0.5, 1.2, 2.4 kg). Florfenicol was administered orally at a dose of 30 mg/kg. The drug was administered eight times at 24 h intervals. The chickens were immunized with SRBC 24 h after administration of the third dose of florfenicol. Florfenicol increased the percentage of CD3+ blood lymphocytes with a corresponding decrease in the percentage of B lymphocytes in birds weighing 0.5 and 2.4 kg. Florfenicol reduced the production of total anti SRBC-haemagglutinins on day 5 after antigen injection in all three body weight groups of the broiler chickens. In conclusion, florfenicol exerted a modulating effect on the immune response of the birds and this should be taken into consideration when using this antibiotic for certain indications.

Abstract

This article discusses some recent developments in the US jurisprudence concerning state immunity. Some lower courts’ decisions handed down earlier suggested a more decisive departure from the rigid interpretation of the Foreign Sovereign Immunity Act (FSIA). If the US Supreme Court had accepted this new jurisprudential trend, it would possibly allow for carving out a partial acceptance of a human rights exception. However, the Supreme Court decided otherwise. In the recently handed-down decision in Germany et al. v. Philipp et al., the Justices rejected any innovations, unequivocally maintained the strict interpretation of FSIA §1603(a)(3), and by their direct reference to the International Court of Justice strengthened the existing status quo in international law as well. This note analyzes this decision’s possible consequences at the domestic and international levels. In conclusion, it seeks to place Germany vs. Philipp in a broader context. It suggests that it possibly reflects more general tendencies in the contemporary US jurisprudence, which can impact both the US domestic legal order and international law.

Abstract

GNSS systems are susceptible to radio interference despite then operating in a spread spectrum. The commerce jammers power up to 2 watts that can block the receiver function at a distance of up to 15 kilometers in free space. Two original methods for GNSS receiver testing were developed. The first method is based on the usage of a GNSS simulator for generation of the satellite signals and a vector signal RF generator for generating different types of interference signals. The second software radio method is based on a software GNSS simulator and a signal processing in Matlab. The receivers were tested for narrowband CW interference, FM modulated signal and chirp jamming signals and scenarios. The signal to noise ratio usually drops down to 27 dBc-Hz while the jamming to signal ratio is different for different types of interference. The chirp signal is very effective. The jammer signal is well propagated in free space while in the real mobile urban and suburban environment it is usually strongly attenuated.

Abstract

Both sarcoidosis and primary Sjögren syndrome (pSS) are multisystem disorders of unknown etiology, which share certain clinical features, making the diff erential diagnosis a real challenge in clinical practice. Several published case reports and case series have posed the question as to whether there is a real association — mimicry between the two diseases or it is just coincidence. We attempt, aft er systematic and comprehensive research of the relevant published literature, to present all those data, clinical or paraclinical, which could be useful in the diagnostic approach and the distinguishment of the two diseases. It is certain that, besides the classic diagnostic methods, emerging is the role of immunology and genetics on this direction, although not established yet.

Abstract

This study aimed to evaluate the nutritional behavior and some immunological criteria (encapsulation index and phenoloxidase – PO activity, the key enzyme for melanization) as well as to study the effect of protein to fat (P : F) diets on hypopharyngeal gland (HPG) protein content. Bees were restricted to consuming specific P : F diets varying in fat ratio under laboratory conditions. These diets included 25 : 1, 10 : 1, 5 : 1 (low-fat diet, LFD); 1 : 1 (equal-fat diet); 1 : 5, 1 : 10 (high-fat diet, HFD), and 1 : 0 (zero-fat diet) as a control. Bees preferred low-fat diets over high-fat diets, where it was 11.27 ± 0.68 μl · day–1 bee in 10 : 1 P : F, while it was 4.99 ± 0.67 μl · day–1 bee in 1 : 10 P : F. However, sucrose consumption was higher in high-fat diets where it was 25.83 ± 1.69 μl · day –1 bee in 10 : 1 P: F, while it was 30.66 ± 0.9 μl · day–1 bee in 1 : 10 P : F. The encapsulation index and phenoloxidase activity of bees were positively linked with the fat level they consumed during all 10 days. The maximum percentage of encapsulation index was 74.6 ± 7.2% in bees fed a high-fat diet, whereas the minimum percentage was 16.5 ± 3.6% in bees which consumed a lowfat diet. Similarly, phenoloxidase activity increased in the haemolymph with increasing fat consumed by bees (0.001 ± 0.0001 and 0.005 ± 0.0003 mM · min –1 · mg –1 at 25 : 1 and 1 : 10 P : F, respectively). The protein content of hypopharyngeal glands in bees which consumed HFD was double that of LFD. Overall results suggest a connection between a fat diet and bee health, indicating that colony losses in some cases can be reduced by providing a certain level of fat supplemental feeding along with sucrose and protein nutrition.

Abstract

Canine parvovirus (CPV) is a single-stranded DNA virus that causes severe and fatal gastrointestinal diseases in dogs. CPV has developed several strategies to evade innate immune response mediated by type I interferons (IFN-I) to achieve a successful infection. The aim of this work was to evaluate the capability of CVP-2c to evade the IFN-I mediated response in infected cells. To establish the role of this response, the gene expression of interferon β (IFNβ), IFIT1, IFIT3, MAVS, and STING were estimated in MDCK cells infected with CPV-2c. Viral replication and gene expression was evaluated by quantitative PCR, also, a treatment with IFN-I (interferon omega) was included to confirm the role of IFN-I during CPV infection. The results revealed that CPV-2c infection stimulates the expression of IFNβ moderately, in these cells. Due to low IFNβ induction, the IFIT1 and IFIT3 expression were also low, and therefore CPV-2c was able to replicate in these cells. However, when the cells were treated with exogenous IFN-I, the IFNβ expression was higher, leading to an increased gene expression of IFIT1 and IFIT3, responsible for antiviral control. The overexpression of these proteins reduced the expression of NS1 and VP2 viral genes and hence viral replication. MAVS and STING expression on infected cells showed a mild increase compared to IFNβ, suggesting that the viral infection could partially modify its expression. All results obtained in this study showed that during CPV-2c infection in MDCK cells, the IFNβ expression was altered since this cytokine is one of the most critical factors for the control and inhibition of viral replication.

Abstract

The aim of the study was to determine the effects of feed addition of LAVIPAN PL5 probiotic preparation containing compositions of microencapsulated lactic acid bacteria ( Leuconostoc mesenteroides, Lactobacillus casei, Lactobacillus plantarum, Pediococcus pentosaceus) on production parameters and post-vaccinal immune response in pigs under field condition. The study was performed on 400 pigs in total and 60 pigs from this group were used to evaluate the effect of the product tested on the post-vaccinal response. The animals were divided into two groups: control group, fed without additive of LAVIPAN PL5 and the study group, receiving LAVIPAN PL5 at doses recommended by manufacturer from weaning to the end of fattening. The following parameters were recorded: main production parameters, including weight gains, fattening time (slaughter age) and animal health status during the study (mortality), and specific humoral post-vaccinal response after vaccination against swine erysipelas. The results indicate that the application of LAVIPAN PL5 had positive influence on the animals` productivity and did not significantly affect the post-vaccinal antibody levels and the development and maintenance of the post-vaccinal response, albeit the levels of antibodies were slightly higher in the animal receiving the test preparation. The higher average daily weight gains (by over 3%) which resulted in a 2 kg higher average weight at slaughter and a reduction of the fattening period by 5 days, undoubtedly contributed to significant economic benefits.

Abstract

The interstitial cells known as telocytes have been described in various organs. Their role in the normal physiology and pathogenesis of numerous diseases is well known. They have been described in the context of various diseases (gallstone disease, endometriosis, uterine myoma, hydronephrosis, myocardial infraction, psoriasis, etc.), while their impact on inflammation, involvement in angiogenesis, and repair highlights their part in local homeostasis. What is known about their relationship with the immune system? Their secretomes, genome, immune profiles, contacts with surrounding cells, and specific loca-lization allow us to give a possible explanation for their involvement in pathological pathways. This review aims to present the roles and features of telocytes in the context of intestinal immunity (the largest in our body), in the spleen, their interactions with immunocytes, and their place in stem cell niches.

Abstract

The outbreak of the war in Ukraine in 2022 resulted in the revival of long-lasting disagreements in Polish-Russian relations. One aspect concerns numerous Russian properties in Warsaw, many abandoned or used for non-public purposes, and the disparity between both States’ properties in the other in this regard. Although the Polish Government has sought to resolve this matter amicably for many years, ultimately several legal proceedings were initiated in Polish courts aimed at recovering some of those premises. Only recently, however, Poland has resolved to employ more decisive steps, including the seizure of the former Soviet residential complex known as Spyville in order to enforce a final judgment. This article sketches the history of the dispute, provides an insight into court proceedings, and discusses the approach of Polish judicial institutions to diplomatic and State immunities.

Abstract

This article presents the results of the research of noiseimmunity of wireless communication systems using signals that are formed on the basis of eight-position quadrature-amplitude modulation (8-QAM) and eight-position amplitude modulation of many components (8-AMMC). The research was conducted using simulation of a wireless communication system, built using a detector, implemented on the basis of a phase locked loop. The influence of phase locked loop parameters on the detection quality of these signals in the condition of the interference in the communication channel was researched, and a comparative analysis of the noise immunity of wireless communication systems using these signals was carried out.

Abstract

Injection of lymphokine activated killer (LAK) cells is known as useful for activation of cellular immune system. Although the effect of LAK cells has been clarified in human or mice, this effect on function of immune cells has not been examined in calves. Healthy ten Holstein calves were injected with the LAK cells 2 days after birth (LAK Group), and another eight calves were observed as controls (Control Group). All calves received the colostrum formulation on the day of birth, and then, were inoculated with a live attenuated vaccine of bovine herpesvirus (BHV)-1 at 2 (the first vaccination) and 6 (the second vaccination) weeks after birth. Peripheral blood of their dam obtained 3 weeks before calving was used for preparation of LAK cells. Blood samples were taken prior to vaccine inoculation and 3 days after the first inoculation, as well as 3 and 6 days after the second vaccination from all calves. Numbers of CD8+ and CD21+ cells increased significantly after the second vaccination in the LAK Group compared with Control Group. The present study suggested the improved effect of injecting LAK cells originated from dams on immune cells function of young calves after BHV-1 live vaccine.

Abstract

To explore the role of Toll-like receptors (TLRs) and interferon (IFN) in the innate immunity against porcine epidemic diarrhea virus (PEDV), we detected the expression of TLR genes in PEDV-infected IPEC-J2 cells by real-time PCR. We also detected the level of interferon α (IFN-α) and interferon γ (IFN-γ) by enzyme-linked immunosorbent assay (ELISA). Results showed that IPEC-J2 cells exhibited a clear pathological change after PEDV infection at 24 h. In addition, TLR7, TLR9 and TLR10 expressions were significantly upregulated in PEDV-infected IPEC-J2 cells at 24 h. Interestingly, the expression patterns of TLR2 and TLR4 were consistent at different stages of PEDV infection. The expression level of TLR3 decreased significantly with the increase of infection time, but the expression levels of TLR5 and TLR8 genes at 6 h and 12 h were significantly lower than those in the control group (p<0.01). There were significant correlations among the expression levels of TLR genes (p<0.05). Cytokine detection showed that the secretion level of IFN-α in the PEDV-infected group was significantly higher than that in the control group (p<0.01), and IFN-γ at 6 h and 12 h after PEDV infection was significantly higher than that in control group (p<0.01). Therefore, our results suggest that PEDV infection can induce innate immune responses in intestinal porcine jejunum epithelial cells, leading to changes in the expression of Toll-like receptors, and can regulate the resistance to virus infection by affecting the release levels of downstream cytokines.

Abstract

Lipopolysaccharide (LPS), a core part of gram-negative bacteria, is crucial for inducing an inflammatory response in living things. In the current study, we used LPS from Salmonella to stimulate chicken macrophages (HD11). Proteomics was used to investigate immune-related proteins and their roles further. Proteomics investigation revealed 31 differential expression proteins (DEPs) after 4 hours of LPS infection. 24 DEPs expressions were up-regulated, while seven were down-regulated. In this investigation, ten DEPs were mainly enriched in S. aureus infection, complement, and coagulation cascades, which were all implicated in the inflammatory response and clearance of foreign pathogens. Notably, complement C3 was shown to be up-regulated in all immune-related pathways, indicating that it is a potential protein in this study. This work contributes to a better understanding and clarification of the processes of Salmonella infection in chickens. It might bring up new possibilities for treating and breeding Salmonella-infected chickens.

Abstract

The present study was aimed to evaluate the growth performance and immune response of three genetic lines of Japanese quails. These lines i.e., selected for 4-week body-weight group (WBS), selected for egg number (EBS), and random-bred control (RBC), were selected for three consecutive generations from a base population of 1125 quails. In total, 2700 four-week-old quails from three selected groups were slaughtered in total of four generations (G0 to G3). Effects of selection and generations as well as their interactions were assessed for growth performance and immune response by applying a two-way analysis of variance. Significant means were compared with Duncan’s Multiple Range Test. The statistical analysis showed a significant effect of selection and generations on most of the growth and immune response parameters. WBS in G3 presented significantly higher values of body weight, weight gain, and FCR than RBC and EBS. FCR was better in WBS during G3 than those of EBS and RBC. However, Livability% was highest in RBC while the lowest was noted in G3 of WBS line. Thymus% and spleen% were higher in EBS as compared to RBC and WBS. RBC presented a better B/S ratio and ND titer than those of EBS and WBS. The decreasing trend of ND titer in both lines of WBS and EBS as compared to RBC suggested a decrease in New Castle disease resistance in progressive generations of selection. It was concluded that selection for body weight and egg number has a positive impact on respective traits but negatively affects the immunity in later generations.

Abstract

The use of lactoferrin (LF) and/or lactobacillus sp. (LB) to improve animal health and production has increased recently. However, information regarding the immune-modulatory role of LB supplementations either alone or in combination with LF in sheep remains unclear. Therefore, the present study was designed to evaluate the immune modulating properties and the antioxidant activity of supplementing commercially available LF and/or LB in healthy lambs. For this reason, twenty-four apparently healthy Ossimi lambs were used. After three weeks of acclimatization, the lambs were randomly allocated to four equal-sized groups and assigned to receive one of the following supplements: LB at a dose of ~ 1 g active ingredient/head (group 1), LF at a dose rate of 0.5 gm /head (group 2), a combination of both treatments using the same dosing regimens (group 3), and (group 4) received only 10 mL of isotonic saline and was considered as a control group. All supplements were given orally twice daily for 30 consecutive days. Blood samples were collected from each lamb before starting the experiment (T0) and two weeks (T15), and four weeks (T30) after giving supplements for hematological examinations, serum biochemical analyses, and RT-PCR assays. Our findings demonstrated that lambs receiving LB showed statistically significant (P<0.05) higher values of total leucocytes, lymphocytes and lysozyme activity than those receiving LF. In contrast, lambs that received LF had significantly (P< 0.05) higher values of serum catalase, nitric oxide and GSH with a significantly lower MDA level compared with those supplemented with LB. A combination of LF and LB supplementation elicited maximal up-regulation of Tollip, TLR4, IL-5, and IL-6 gene expression compared with other groups. The results suggest that bovine LF and or LB could be used as useful nutritional supplements to support the immune system in healthy lambs.

Abstract

The aim of the experiment was to examine the effect of a diet enriched with Lactobacillus plantarum and/or β-glucan on the immune parameters in the juvenile tench (Tinca tinca). Fish were fed for 14 days different diets (phase 1 of the experiment), a dry commercial starter feed in the control group or the same feed supplemented with: 1% β-1,3/1,6-glucan in group G, 108 cfu L. plantarum g-1 in group L, 1% β-1,3/1,6-glucan + 108 cfu L. plantarum g-1 in group G+L. During consecutive 14 days all fish were fed the commercial feed alone (phase 2). The stimulating effects of the tested preparations was evaluated twice, at the end of each experimental phase. Dietary supplementation of β-1,3/1,6-glucan considerably improved the humoral innate immune response (activity of lysozyme and total Ig) and the pinocytotic activity of phagocytes. Supplement of L. plantarum improved the ability of the head kidney phagocytes (RBA) to carry out oxygen burst in L and G+L groups. A similar effect was observed for the killing activity of phagocytes (PKA) from the head kidney after the stimulation of A. hydrophila, and the effect persisted for two weeks after the commercial feed regime was resumed. A significant increase in the pro- liferative activity of B lymphocytes originating from the head kidney was observed in groups L and G+L. The study has revealed that the addition of the tested G+L synbiotic to dry diet stimulates the innate immune response mechanisms in the juvenile tench.

Abstract

Plant viruses cause crop losses in agronomically and economically important crops, making global food security a challenge. Although traditional plant breeding has been effective in controlling plant viral diseases, it is unlikely to solve the problems associated with the frequent emergence of new and more virulent virus species or strains. As a result, there is an urgent need to develop alternative virus control strategies that can be used to more easily contain viral diseases. A better understanding of plant defence mechanisms will open up new avenues for research into plant- pathogen interactions and the development of broad-spectrum virus resistance.
The scientific literature was evaluated and structured in this review, and the results of the reliability of the methods of analysis used were filtered. As a result, we described the molecular mechanisms by which viruses interact with host plant cells.
To develop an effective strategy for the control of plant pathogens with a significant intensity on the agricultural market, clear and standardised recommendations are required. The current review will provide key insights into the molecular underpinnings underlying the coordination of plant disease resistance, such as main classes of resistance genes, RNA interference, and the RNA-mediated adaptive immune system of bacteria and archaea – clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated Cas proteins – CRISPR/Cas.
Future issues related to resistance to plant viral diseases will largely depend on integrated research to transfer fundamental knowledge to applied problems, bridging the gap between laboratory and field work.

Abstract

In the skeptical tradition self-consciousness was transparent and it served as a basis for expressing doubts and developing arguments leading to certainty. After the linguistic and naturalistic turns, contemporary philosophy developed skeptical arguments against certainty and epistemic priority of the data of self-consciousness (both reflective and pre-reflective). Self-reflection reports on the stream of consciousness ex post, but the reports are meager and dependent on subject’s conceptual scheme, while the pre- -reflective data is unclear. Two contemporary skeptical hypotheses have been developed: H. Putnam’s content externalism hypothesis and so-called Kripkenstein’s quus hypothesis. I put forth the question what kind of self is immune to erroneous misidentification. The immunity seems to be limited to the contentless self, reducible to the pre-discursive feeling of one’s own existence. There is no guarantee that any content whatsoever can be attributed to self without error. I cannot negate that I exist any more than I can negate that something external exists, but any description of either is fallible. So the content of self-consciousness is not in an epistemically better position than the content of external perception.

Abstract

Prolonged exposure to stress may cause adverse effects on animal physiology. It is especially important during the gestation period as female physiology can affect the unborn offspring in the form of prenatal stress. Intensive pig farming industry developed gestation crates that enable to keep sows during gestation period in small stalls which do not allow animals to move freely for a maximum of 4 weeks after successful insemination (Council Directive 2008/120/EC). Although these crates have production advantages, many health and welfare issues have been raised recently. In this study we tested to what extent the lack of movement of sows kept in the gestation crates had an impact on some blood and saliva constituents of new-born piglets. In total, the samples were collected from 80 piglets when they were 3, 7 and 21 days of age and tested for cortisol levels in blood and saliva, acute phase proteins (amyloid A, C-reactive protein, haptoglobin) and lymphocytes proliferation index (in response to ConA, PHA and PWM). 40 piglets were from sows kept in free movement housing (FM group) from day 1 to day 100 of pregnancy and forty piglets were from sows in the movement restriction group (MR), in which the sows were kept in crates just allowing them to stand up and lie down from day 1 to day 100 of the pregnancy (research was conducted before the implementation Directive 2008/120/EC i.e. January 1,2013). The results of the study showed that the piglets delivered by sows kept under movement restriction conditions exhibited higher cortisol and acute phase protein levels as well as a lower lymphocytes proliferation index. This suggests that lack of movement in sows during the gestation period influences piglets’ physiology and indicates that the piglets are suffering from prenatal stress caused by insufficient housing conditions of their mothers potentially leading to poor health and welfare of their offspring.

Abstract

From the announcement of the Independence by Ukraine in 1991 and signing many bilateral agreements with the Russian Federation on the state border, the joint use of the Azov Sea and the Kerch Strait, until the annexation of the Crimea in 2014, both parties did not manage to carry out maritime distractions on these waters, due to different positions. After the annexation of the Crimea, the situation became beneficial for FR, because in its hands there was a Kerch Strait and the Russian side started the economic blockage of Ukrainian ports on the Azov Sea. The Ukrainian site, wanting to increase the number of its ships in the Berdyansk base located on this sea, decided to move through the sea team consisting of 2 artillery cutters and a subsidiary (tug) belonging to the Navy of Ukraine. During the approach to Kerch Strait, an incident occurred on November 25, 2018 between Ukrainian ships and the ships of the border service of the Federal Security Service (shelling, detention of the ships, arrests and putting their crews before the court). The above action was inconsistent with international law violating Articles 32, 58, 95 and 96 of the Convention on the Law of the Sea from 1982. The International Tribunal for the Law of the Sea in its judgement ordered RF to give back the ships and send seamen to Ukraine. The warrant was performed partially: ships were returned, while seafarers returned as part of the exchange of prisoners between the parties. Currently, Russia continues to run the economic blockage of the Ukrainian ports, closing (due to military exercises) approaches to the Kerch Strait for ships and vessels of other countries. The current situation indicates that Russia implements its geopolitical goals and disregards the provisions of international law of the sea.

Abstract

Immunoaffinity chromatography (IAC) is a fundamental isolation and purification tool which is incorporated in a substantial range of therapeutic and diagnostic applications. This study has reappraised the usefulness of immunoaffinity chromatography for the purification of polyclonal antibodies. Protein A based IAC is a convenient and reliable method for purification of IgG, from hyperimmunesera (HIS) raised in experimental animals such as rabbits, guinea pigs and mice to be utilized in pharmaceutics and diagnostics. The 146S fraction of Foot and Mouth Disease virus (FMDV) TCID50=10 5.6 was cultured on a baby hamster kidney cell line 21 (BHK-21), concentrated using salt precipitation method using PEG 6000, purified by size exclusion chromatography (SEC) using Sepharose-30 at 254nm absorbance. Purification of 146S FMDV was analyzed using 12% SDS-PAGE which provided two bands of light and heavy chains. The alum-based vaccine, consisting of ≥10μg of 146S FMDV, was applied in 10 male rabbits and 10 male guinea pigs and two animals of each group were taken as a negative control. The titer of serum was calculated using virus neutralization test. A Protein-A kit (Thermo scientific- 44667, 0528.2) was used to purify HIS raised against 146S FMDV and validated using 12% SDS PAGE in reducing condition. The data demonstrate that protein-A affinity chromatography is an efficient tool for the purification of antibodies from hyper-immune sera raised against 146S FMDV and can be used for the production of diagnostic kits e.g. Enzyme linked immuno-sorbent assay (ELISA) and radioimmunoassay.
